Fig. S2. Additional examples for the differential effects of Nkx6.1 overexpression on Olig2 and Mnr2/Hb9 gene expression at different developmental stages. E2 chicken spinal cords were electroporated with RCASBP-Nkx6.1 retroviral expression vector, and then were processed two days (at E4, A-C) or four days (at E6, D-I) later for gene expression studies by immunofluorescence with anti-Nkx6.1 (A,D,G), anti-Olig2 (B,E,H), anti-Mnr2/Hb9 (C,F,I) antibodies. Two days after Nkx6.1 electroporation, Olig2 and Mnr2/Hb9 were induced in the dorsal spinal cord (A-C). However, 4 days after electroporation, the ectopic expression of Olig2, but not Mnr2/Hb9, was absent from the dorsal spinal cord. Moreover, the endogenous expression of Olig2 was inhibited when Nkx6.1 is overexpressed in the ventral spinal cords (D-F), but not when ectopic expression of Nkx6.1 only occurred in the dorsal spinal cords (G-I). Induction of gene expression is indicated by arrows, whereas inhibition by arrowhead.
