Fig. S1. Epithelial adhesion molecules in PBKO pancreata. E11.5 wild-type (A-D) and PBKO (A9-D9) dorsal pancreata were analyzed by confocal immunofluorescence to detect b-catenin, plakoglobin and E-cadherin expression. Whereas plakoglobin expression is barely detectable in wild-type pancreas (B), it is dramatically upregulated in b-catenin-deficient cells (B9). Expression of E-cadherin, the major cadherin in the developing pancreas (Dahl et al., 1996), is indistinguishable between wild type and PBKO (C,C9).
Fig. S2. Proliferation of differentiated pancreatic cells. BrdU-labeled E15.5 pancreata (pregnant females injected 1 hour prior to harvest, 50 mg/kg) were analyzed by confocal immunofluorescence to detect co-localization of BrdU and insulin (A), amylase (B) or DBA (C), which marks ducts and progenitor cells at this stage (Kobayashi et al., 2002). The BrdU-labeling index for each cell type is indicated (two E15.5 littermates analyzed, ³13 fields per marker, >350 cells counted per marker).
