Movie 1. Time-lapse movie of representative Cdx2 mutant (left) and non-mutant littermate (right). Cdx2 mutants undergo compaction, cavitation and blastocoel expansion like non-mutant littermates, but the blastocoel is not maintained, the TE becomes rough and mutants eventually collapse. A total of three mutants and 18 non-mutants recorded exhibited similar phenotypes.
Fig. S1. Cdx2 transcript expression markes the TE precursors and their derivatives. (A-E) Fluorescent in situ hybridization of Cdx2 antisense RNA probe. Each panel is a composite of DIC and fluorescent images from a single optical section. (A) 3.25 dpc early blastocyst (n=1). (B) 3.5 dpc expanded blastocyst (n=4). (C) 4.5 dpc implanting blastocyst (n=2). (D) 4.75 dpc (n=2). (E) 5.5 dpc (n=2). White arrowheads (C,D) demarcate Cdx2 expression domain in polar TE, bracket indicates ~4 cell rows in ExE where Cdx2 is expressed. Scale bars: 15 mm in A-C; 30 mm in D; 20 mm E.
