Fig. S1. lacZ expression was not detected in proliferating precursors. Pregnant mice were injected at E10.5 intraperitoneally with 100 mg/g body weight of BrdU, and embryos were collected 1 hour after the injection. Transverse section of an embryo carrying Tg4 was immunostained with antibodies against BrdU (red) and b-gal (greeen). Confocal images did not show any overlap of the two colors. No overlap of the two colors was also observed for embryos carrying Tg2 (data not shown). Scale bar: 50 mm.
Fig. S2. lacZ expression of Mbh1/lacZ transgenic embryos. Dorsal views and transverse sections of representative b-gal+ embryos carrying Tg7 (A,B), Tg14 (C,D), Tg15 (E,F) and Tg18 (G,H). An arrowhead indicates the b-gal+ somite. None of embryos carrying Tg16 and Tg17 expressed lacZ in the spinal cord (I,J). No expression of lacZ was also confirmed by sectioning (data not shown). Scale bars: in A, 100 mm for A,C,E,I,J; in B, 100 mm for B,D,F,H; in G, 100 mm.
Fig. S3. Mbh1 expression was lost in the Math1–/– spinal cord at E10.5. Transverse sections at brachial levels of Math1+/+ (A,C) and Math1–/– (B,D) mouse embryos were hybridized with antisense cRNA probes for Math1 (A,B) and Mbh1 (C,D). Loss of Mbh1 expression was observed through all axial levels of the spinal cord in all Math1–/– embryos (n=3). Scale bar: 100 mm.
Fig. S4. Mbh1 expression was not detected in the Math1–/– spinal cord even at later stages. Transverse sections at brachial levels of E14.5 Math1+/+ (A) Math1–/– (B) mouse embryos were hybridized with antisense cRNA probes for Math1 (A) and Mbh1 (C). Loss of Mbh1 expression was observed through all axial levels of the spinal cord in all Math1–/– embryos (n=3). As Math1 expression ceased in the spinal cord at E14.5, lack of Math1 expression in the Math1–/– embryos was confirmed using hindbrain sections (data not shown). Scale bar: 100 mm.
