Fig. S1. E4.5 (HH25) gonads from embryos infected with RCAS-Pitx2 or RCAS-AP and analysed for the expression of Pitx2 and Bmp7. At this stage, Bmp7 is normally expressed in the left gonad, as is Pitx2. The misexpression of Pitx2 to the right induces Bmp7 expression in the right gonad. (A) The RCAS infection was carried out at HH8-10. Whole-mount in situ hybridisation for Pitx2 and Bmp7 on the posterior and, respectively, anterior portion of a gonad from (top row) the control experiments (7/8 as shown; one expressed Pitx2 bilaterally and was discarded) or (bottom row) from the RCAS-Pitx2a experiments (10/10 as shown). (B) The experiment was performed by co-electroporation of the virus DNA and a GFP plasmid at HH15-16. Only the gonads showing high levels of GFP expression at E4.5 were retained. The in situ hybridisation for Pitx2 and Bmp7 on adjacent sections of a gonad from (top row) the control experiment or (bottom row) the RCAS-Pitx2c experiment indicates that also late misexpression of Pitx2 to the right gonad is sufficient to induce Bmp7 expression (three gonads analysed per experiment).
