  Fig. S1. ECFP protein tagged with three classical basic nuclear localisation sequences (ECFP-NLS) does not accumulate in the nucleus until after the MBT. (A) Animal pole blastomeres derived from embryos injected with RNA encoding ECFP-NLS together with ECFP-GPI membrane marker (right-hand panel), or VNm9-Smad4 and VC155-Smad2 (middle panel), were treated with Activin at early blastula stage 7 and then cultured until the indicated stages. Note that nuclear accumulation of Smad complexes and of ECFP-NLS does not occur until the MBT. (B) Cells derived and cultured as in A but not treated with Activin.
