Fig. S1. Zygotic expression of MRG-1. (A-C) To determine when after fertilization and in which cells mrg-1 is expressed from the embryonic genome, M+Z- fertile mrg-1(qa6200) hermaphrodites (lacking a maternal load of protein) were mated with wild-type males, and their M-Z+ mrg-1/+ offspring were stained with rabbit anti-MRG-1 (red) and DAPI (blue). Zygotically produced MRG-1 was not detected in early embryos (circled by dotted lines in A), became detectable in all nuclei by the ∼100-cell stage (arrow in A and B), and was enriched in the PGCs (arrowheads in C) relative to somatic cells by the 2- to 3-fold stage. Scale bars: 10 μm.
Fig. S2. MRG-1 is not required for the histone methyltransferase activity of MES-4 or of the MES-2/MES-3/MES-6 complex. (A-F) Two-cell embryos stained with mouse PA3 (A,D; red in merge) and rabbit anti-H3K36me2 (B,E; green in merge). (A-C) Wild-type embryo. (D-F) mrg-1(qa6200) M-Z- embryo. Arrows point to X chromosomes. (G-J) Two-cell embryos stained with mouse PA3 (G,I) and rabbit anti-H3K27me3 (H,J). (G,H) Wild-type embryo. (I,J) mrg-1(qa6200) M-Z- embryo. Embryos are approximately 50 μm in length.
