Fig. S1. Expression of Xc-Myc-1 and Xc-Myc-2 during Xenopus development. qRT-PCR analysis of Xenopus embryos at the indicated stages of development for the expression of Xc-Myc-1 (red bars) and Xc-Myc-2 (blue bars). Expression was relative to that found in unfertilized embryos.
Fig. S2. Knockdown of Xc-Myc does not cause obvious defects in early Xenopus development. Embryos injected with the Xc-Myc morpholino (Mo) at the one-cell stage were allowed to develop to the indicated stages. At these intervals, embryos were photographed and analyzed for possible phenotypes. No significant external differences in the development of control or Xc-Myc morpholino-injected embryos were detected at these stages.
Fig. S3. Knockdown of Xc-Myc compromises XSlug and XTwist expression in the neural crest, which is rescued by ectopic Slug or Twist expression. Whole-mount in situ hybridization of embryos injected in one blastomere at the two-cell stage with Xc-Myc morpholino (Mo) alone or when co-injected with the Xc-Myc morpholino and Slug, Twist or Xc-Myc-Mut mRNA. Following fixation, the embryos were analyzed for XSlug or XTwist expression. Injected Slug or Twist mRNA was not detected above background by the in situ analyses.
