Fig. S1. Upregulated expression of Wnt genes in Cre-adenovirus-infected Ptch1c/c primary chondrocytes. Quantitative RT-PCR was performed using RNA isolated from primary Ptch1c/c chondrocytes that had been infected with or without Cre-adenovirus. Wnt1, Wnt2, Wnt3a, Wnt4, Wnt6, Wnt7a, Wnt8, Wnt8b, Wnt11, Wnt15 and Wnt16 were upregulated in the Cre-adenovirus-infected samples.
Fig. S2. Hedgehog signaling does not stimulate hypertrophy in the presence of PTHrP signaling. Quantitative RT-PCR was performed using RNA isolated from primary chondrocytes. Expression of Ihh, ColX, Bmp6 and PTHR was decreased in Shh-treated primary chondrocytes compared with untreated samples, possibly because of strong PTHrP signaling in these cells. Dkk1 and Noggin treatment further inhibited ColX expression. However, as Wnt and Bmp signaling may also interact with PTHrP signaling, the effect of Dkk1 and Noggin treatment was more complicated in the primary costal chondrocytes.
